NARRATIVE:

The overall focus of our research is to determine the underlying metabolic alterations in pathological states. Specifically, our research centers around utilizing mass spectrometry-based lipidomic and metabolomics approaches to better understand diabetes-induced alterations in the retina and testing the contributions of these observations to diabetic retinopathy pathogenesis. One of our findings is an elevation of glucosylceramide, a sphingolipid, in the diabetic retina. Glucosylceramide synthase catalyzes the formation of glucosylceramide and is the first committed step in the formation of several higher order glycosphingolipids, including gangliosides and globosides. Our continued efforts are to understand the underlying mechanisms for diabetes-induced modulation of glycosphingolipid metabolism and to ascertain the biochemical and biophysical roles of these lipids. Therapeutic and transgenic approaches are also being utilized to assess the roles of these glycosphingolipids in normal retinal homeostasis and diabetic retinopathy. In collaboration with other researchers, similar metabolomics and lipidomics approaches are also being applied to several diseases, including diabetes and various cancers. These team projects are focused on delineating alterations that occur within clinical specimens.
